All cells were cultured in a final volume of 200 µl in the presence of 1 × 104 irradiated peripheral mononuclear cells as antigen-presenting cells. All tests were conducted in triplicate. Cell cultures were then incubated at 37°C for 4 days and supernatants were obtained for cytokine measurements before AZD4547 cell line being pulsed with 1 µCi [3H]-thymidine per well for the final 18 h of incubation. Plates were harvested onto nylon filters using the Betaplate system and radioactivity was quantified using a Betaplate counter.
